It was the first time that the number of infections topped 20,000 in Tokyo.
The daily tally is about 4,000 more than the previous high of 17,631, which was recorded last Friday. It is also around 1.5 times the daily figure reported a week ago.
Tokyo officials say 30 patients are in serious condition, based on the criteria set by the Metropolitan Government. The number is up one from Tuesday.
The occupancy rate of hospital beds for coronavirus patients in Tokyo is 51.4 percent as of Wednesday, up 0.7 percentage point from Tuesday.
